چکیده

A Ca2+ indicator has been synthesized and characterized which can be used to monitor rapid changes in the free Ca2+ concentration ([Ca2+]) immediately adjacent to cell membranes. This indicator, referred to as C18-Fura-2, consists of a Fura-2 molecule conjugated to a lipophilic alkyl chain which will insert into cell membranes. When associated with cell membranes in low concentrations, C18-Fura-2 exhibits an excitation spectrum with a large Stokes shift and a single isobestic point, thus [Ca2+] can be calculated ratiometrically. The apparent Ca2+ dissociation constant of cell-associated C18-Fura-2 is around 150 nM. C18-Fura-2 orients in the cell membrane so that the fluorophore is facing the side to which it was applied. C18-Fura-2 was used to record rapid changes in intracellular [Ca2+] which occurred in response to membrane depolarization in isolated smooth muscle cells. The initial rise of the [Ca2+] transient reported by C18-Fura-2 was four to six times faster than the rise of the [Ca2+] transient reported by cytosolic Fura-2. This result suggests that C18-Fura-2 was located at the plasma membrane near sites of Ca2+ influx and indicates that membrane-associated Ca2+ indicators can be used to detect rapid, localized changes in [Ca2+] which are obscured in signals recorded using water-soluble, bulk cytosolic fluorescent Ca2+ indicators.

Modification of Nifedipine Inhibitory Effect on Calcium Spike and L-Type Calcium Current by Ethanol in F1 Neuron of Helix aspersa

There is strong evidence demonstrating that nifedipine dissolved in ethanol selectively inhibits only L-type Ca2+ current. In addition, acute ethanol exposure reduces voltage-dependent calcium currents. In the present study, we investigated the antagonistic effect of fixed concentration of nifedipine dissolved in different concentration of ethanol on L-type Ca2+ current. In a Na+-K+ free soluti...

Measurement of intrasynaptosomal free calcium by using the fluorescent indicator quin-2.

The recently synthesized calcium indicator quin -2 was incorporated into synaptosomes from guinea-pig cerebral cortex following uptake and internal hydrolysis of quin -2 tetra-acetoxymethyl ester. Incubation in physiological media containing 1 mM- or 2 mM-CaCl2 led to equilibrium cytosolic ionized calcium concentrations of 85 +/- 10 nM and 205 +/- 5 nM respectively (mean +/- S.E.M. from eight a...

O-31: Mifepristone Acts as Progesterone Antagonistof Non-Genomic Responses but InhibitsPhytohemagglutinin Induced Proliferationin Human T Cells

Background: Progesterone is an endogenous immunomodulator that suppresses T cell activation during pregnancy. The stimulation of membrane progesterone receptors (mPRs) would seem to be the cause of rapid non-genomic responses in human peripheral T cells, such as an elevation of intracellular calcium ([Ca2+] i) and decreased intracellular pH (pHi). Mifepristoneimmune cells compared with progeste...

Insulin increases near-membrane but not global Ca2+ in isolated skeletal muscle.

It has long been debated whether changes in Ca2+ are involved in insulin-stimulated glucose uptake in skeletal muscle. We have now investigated the effect of insulin on the global free myoplasmic Ca2+ concentration and the near-membrane free Ca2+ concentration ([Ca2+]mem) in intact, single skeletal muscle fibers from mice by using fluorescent Ca2+ indicators. Insulin has no effect on the global...

Cu(II) Ion- Selective Membrane Electrode Based on bis(N-salicylidene-3,3'-diamino diphenyl) sulfone

In order to study the Cu(II) ion selective electrode, an optimized membrane was prepared by mixing of 30 mg of PVC powdered, 5 mg of considered ionophore, 60 mg of plasticizer and 5 mg of additive. The sensor showed a near Nernstian response for Cu(II) ions over a concentration range from 1.0×0-6 –1.0×0-2 M with a slope of 28.90.5 mV per concentration decade with a working pH range of 3.5-6.3. ...
